I have one! Somewhere in your Startup an Assign is made to this directory.
Rename it, reboot, and see where (if) your startup fails. If it doesn't fail on startup, you will probably eventually see a requester asking you to "Insert Volume dir:". Then you will know which program is referring to it, and you will have to change something in that program's prefs.
Anyway, you should then be able to delete it (after the reboot), but I would wait until I figured out what program is refering to it.
